Drug developers increasingly require faster compound screens to reduce time-to-market. With the BioTrove RapidFire™ XC-MS System, in vitro ADME assays can be performed using a time-tested, reliable tool—the mass spectrometer, at previously unachievable throughputs.

An 8-point enzyme timecourse in triplicate was run in 2.6 minutes. The substrate and product signals in each of the 24 samples are shown in the above traces.

The RapidFire™ XC-MS system's patented chromatography process cleans and purifies test compounds before introduction into the mass spectrometer. The RapidFire™ XC-MS system operates at high speed, between 5 and 15 seconds per sample, making it ideal for the screening of large numbers of test compounds.

BioTrove has already developed assays for screening the specific activity of compounds against individual P450 isoforms. The assays are compatible with liver microsome preparations as well as recombinant enzymes. Mass spectrometer incompatible salts and buffers are separated from the analytes of interest using rapid chromatography prior to MS analysis.

These drug-drug interaction assays can be run with throughputs of 8-10 seconds per sample or less. This is equivalent to one hundred 96-wells plates per day!

BioTrove scientists are developing additional assays for the RapidFire™ XC-MS System. Additionally, our scientists can partner with pharmaceutical researchers to develop proprietary assays for specific screens.
